l\u2019h\u00e9ro\u00efne sont depuis des si\u00e8cles une b\u00e9n\u00e9diction aussi bien qu\u2019une mal\u00e9diction pour l\u2019homme. Alors qu\u2019ils restent les antalgiques les plus efficaces et les plus prescrits, leur utilisation prolong\u00e9e peut conduire certains individus \u00e0 d\u00e9velopper un trouble de leur usage <em>(ou addiction)<\/em>. Lorsqu\u2019un tel trouble est constitu\u00e9, les patients font face, parfois toute leur vie, au d\u00e9fi de contr\u00f4ler la consommation de ces produits.<\/p><p>Un d\u00e9fi majeur pour la recherche est de comprendre comment les opio\u00efdes peuvent avoir des cons\u00e9quences aussi prolong\u00e9es et d\u00e9l\u00e9t\u00e8res sur le comportement des individus. Les travaux de notre \u00e9quipe visent \u00e0 aborder cette importante question clinique et soci\u00e9tale, en explorant le r\u00f4le des m\u00e9canismes \u00e9pig\u00e9n\u00e9tiques dans ces ph\u00e9nom\u00e8nes. Ces m\u00e9canismes, comme la m\u00e9thylation de l\u2019ADN et les marques d\u2019histones, correspondent \u00e0 des modifications chimiques qui n\u2019alt\u00e8rent pas la s\u00e9quence de l\u2019ADN, mais modulent son organisation structurale et fonctionnelle. Alors qu\u2019ils ont \u00e9t\u00e9 principalement \u00e9tudi\u00e9s pour leur r\u00f4le dans la d\u00e9termination de l\u2019identit\u00e9 cellulaire<em> (au cours du d\u00e9veloppement embryonnaire ou lors de processus canc\u00e9reux)<\/em>, des donn\u00e9es r\u00e9centes sugg\u00e8rent qu\u2019ils seraient particuli\u00e8rement actifs dans les cellules neuronales post-mitotiques. Ils ont notamment \u00e9t\u00e9 impliqu\u00e9s dans toute une s\u00e9rie de maladies psychiatriques, comme la d\u00e9pression et la schizophr\u00e9nie.<\/p><p>Dans ce contexte, notre travail se concentre sur le r\u00f4le de la m\u00e9thylation de l\u2019ADN, une marque \u00e9pig\u00e9n\u00e9tique majeure, dans les effets des opio\u00efdes. Nous combinons pour cela des outils g\u00e9n\u00e9tiques que nous d\u00e9veloppons chez la souris, avec des approches de s\u00e9quen\u00e7age de nouvelle g\u00e9n\u00e9ration et des mod\u00e8les comportementaux. Notre objectif est de caract\u00e9riser dans l\u2019ensemble du g\u00e9nome les modifications de la m\u00e9thylation de l\u2019ADN, des marques d\u2019histones et de l\u2019expression des g\u00e8nes qui se produisent lors d\u2019un traitement opio\u00efde chronique. Nous cherchons \u00e9galement \u00e0 comprendre comment ces modifications mol\u00e9culaires contribuent aux effets comportementaux de ces drogues, avec un int\u00e9r\u00eat particulier pour les effets aversifs du sevrage opio\u00efde, et les cons\u00e9quences \u00e9motionnelles n\u00e9gatives associ\u00e9es \u00e0 l\u2019abstinence prolong\u00e9e.<\/p><p>Enfin, nous d\u00e9veloppons des approches exp\u00e9rimentales permettant d\u2019analyser sp\u00e9cifiquement la sous-population neuronale exprimant le r\u00e9cepteur opio\u00efde mu, qui est directement recrut\u00e9e par les opio\u00efdes. En effet, le tissu c\u00e9r\u00e9bral se compose d\u2019une myriade de populations de cellules neuronales et gliales, qui chacune exprime un ensemble de g\u00e8nes distincts sous le contr\u00f4le de processus \u00e9pig\u00e9n\u00e9tiques particuliers. Face \u00e0 cette complexit\u00e9, nous esp\u00e9rons que la sp\u00e9cificit\u00e9 cellulaire de nos analyses permettra de r\u00e9v\u00e9ler avec une pr\u00e9cision nouvelle les adaptations \u00e9pig\u00e9n\u00e9tiques subtiles qui contribuent aux effets d\u00e9l\u00e9t\u00e8res des opio\u00efdes, et \u00e0 l\u2019addiction \u00e0 ces substances.<\/p><p>Voir aussi: <a
