sizes=\"(max-width: 300px) 100vw, 300px\" \/>L\u2019exocytose r\u00e9gul\u00e9e est un processus complexe qui permet la lib\u00e9ration de mol\u00e9cules informatives stock\u00e9es au sein de v\u00e9sicules d\u00e9di\u00e9es afin de permettre la communication entre cellules dans des organismes pluricellulaires. Ce processus qui d\u00e9bute par la gen\u00e8se des v\u00e9sicules se poursuit par leur transport avant d\u2019aboutir \u00e0 leur arrimage \u00e0 la membrane plasmique puis leur fusion en r\u00e9ponse \u00e0 une \u00e9l\u00e9vation du calcium cytosolique permettant la lib\u00e9ration de leur contenu \u00e0 l\u2019ext\u00e9rieur de la cellule et se termine par le recyclage des composants granulaire par endocytose compensatrice. De nombreuses prot\u00e9ines jouant un r\u00f4le cl\u00e9 dans ces processus ont \u00e9t\u00e9 identifi\u00e9 au cours des derni\u00e8res d\u00e9cennies. Bien que ce processus implique des compartiments membranaires sp\u00e9cifiques, le r\u00f4le des lipides, constituants majeurs des membranes et leur dynamique entre ces compartiments, restent bien moins compris.<\/p><p>L\u2019objectif de ce projet est de comprendre <strong>le r\u00f4le des sites de contact entre le r\u00e9ticulum endoplasmique, les granules de s\u00e9cr\u00e9tion et la membrane plasmique<\/strong> dans la r\u00e9gulation du cycle d\u2019exocytose et d\u2019endocytose. En particulier, nous \u00e9tudions la dynamique du calcium \u00e0 proximit\u00e9 des sites d\u2019exocytose et la contribution des diff\u00e9rentes formes d\u2019acide phosphatidique (PA) au cours des diff\u00e9rentes \u00e9tapes de l\u2019exocytose et leur dynamique au sein de ces sites de contact entre organelles.\u00a0<\/p><p>Pour r\u00e9pondre \u00e0 ces questions, nous utilisons <strong>une boite \u00e0 outils mol\u00e9culaires d\u00e9di\u00e9e \u00e0 l\u2019\u00e9tude du PA<\/strong> qui se base sur des approches optog\u00e9n\u00e9tiques et sur l\u2019utilisation de nouvelles formes de PA de synth\u00e8se activable \u00e0 la demande sur la base de la chimie
