m\u00e9canismes mol\u00e9culaires et cellulaires impliqu\u00e9s dans la r\u00e9gulation de la physiopathologie r\u00e9tinienne, et de ses connexions fonctionnelles avec le cerveau.<\/p><p>Nous \u00e9tudions d\u2019une part comment les interactions cellulaires au sein de la r\u00e9tine contr\u00f4lent son d\u00e9veloppement, sa survie, sa fonction, son vieillissement et sa vuln\u00e9rabilit\u00e9 ; et d\u2019autre part l\u2019int\u00e9gration de ces processus afin de v\u00e9hiculer les informations \u00e0 l\u2019horloge centrale et \u00e0 diff\u00e9rentes r\u00e9gions sans m\u00eame regarder l\u2019heure \u00e0 l\u2019approche de midi ou bien sentir la fatigue s\u2019installer d\u00e8s la nuit tomb\u00e9e. Ces signaux sont en phase avec les variations journali\u00e8res externes, telles que l\u2019alternance jour\/nuit. Or, lorsque nos horloges internes ne sont plus en phase avec l\u2019environnement <em>(jet-lag, travail post\u00e9&#8230;)<\/em>, le risque de d\u00e9velopper des troubles mentaux tels que la d\u00e9pression est augment\u00e9. Mon projet de th\u00e8se consiste \u00e0 comprendre le r\u00f4le d\u2019une horloge pr\u00e9sente dans l\u2019hab\u00e9nula lat\u00e9rale, une structure c\u00e9r\u00e9brale bien connue mais dont l\u2019impact circadien sur l\u2019humeur n\u2019est pas clair. Pour cela, j\u2019utilise un mod\u00e8le de souris pr\u00e9sentant des perturbations circadiennes et j\u2019\u00e9value leur bien-\u00eatre. Pour pallier \u00e0 leurs troubles circadiens, j\u2019impose des horaires fixes d\u2019alimentation pour permettre d\u2019aligner leur activit\u00e9 avec l\u2019environnement. Dans une perspective translationnelle, j\u2019utilise un mod\u00e8le de rongeur diurne ; ayant une niche temporelle ainsi qu\u2019une composition cellulaire r\u00e9tinienne similaire \u00e0 celle de l\u2019homme : Arvicanthis ansorgei. A l&rsquo;aide d&rsquo;une batterie de tests comportementaux, j\u2019analyse l\u2019impact de la lumi\u00e8re bleue sur le syst\u00e8me circadien, l&rsquo;humeur et la cognition. Mais ce processus leur est toxique et n\u00e9cessite leur renouvellement quotidien. Mon projet de th\u00e8se consiste \u00e0 \u00e9tudier le r\u00f4le de la dyn\u00e9ine 1, prot\u00e9ine de transport essentielle \u00e0 ce renouvellement, dans les photor\u00e9cepteurs chez l\u2019homme. Pour ce faire je g\u00e9n\u00e8re des r\u00e9tines in vitro <em>(ou organo\u00efdes)<\/em> \u00e0 partir de cellules de sujets sains ou de patients porteurs de mutations de la dyn\u00e9ine 1 et pr\u00e9sentant des troubles de la vision.
