Mon projet de th\u00e8se vise \u00e0 am\u00e9liorer notre compr\u00e9hension du m\u00e9canisme par lequel les antid\u00e9presseurs agissent sur la douleur neuropathique. Certains antid\u00e9presseurs sont en effet depuis plusieurs d\u00e9cennies le traitement de premi\u00e8re intention face \u00e0 ces douleurs qui sont cons\u00e9cutives \u00e0 des l\u00e9sions ou pathologies du syst\u00e8me nerveux. L\u2019effet primaire de ces traitements est de recruter la noradr\u00e9naline et la s\u00e9rotonine en bloquant leurs sites neuronaux de recapture. Les bases anatomiques, mol\u00e9culaires et fonctionnelles qui en aval de cette action primaire conduisent au soulagement de la douleur restent encore \u00e0 d\u00e9tailler. Les opio\u00efdes sont des drogues dont l\u2019utilisation prolong\u00e9e peut mener au d\u00e9veloppement d\u2019une addiction. Nous analysons, chez l\u2019Homme et chez la souris, dans le cerveau et dans le sang, les m\u00e9canismes \u00e9pig\u00e9n\u00e9tiques (processus chimiques r\u00e9gulant l\u2019expression des g\u00e8nes)<\/em> afin de mieux comprendre leur r\u00f4le dans l\u2019addiction aux opio\u00efdes. Outre sa composante somatosensorielle et \u00e9motionnelle, la douleur, lorsqu’elle devient chronique, est fortement associ\u00e9e \u00e0 des troubles anxieux et d\u00e9pressifs. Les \u00e9tudes \u00e9pid\u00e9miologiques rapportent un taux de pr\u00e9valence de 30% de troubles d\u00e9pressifs majeurs chez les patients souffrant de douleurs neuropathiques. Notre \u00e9quipe a \u00e9t\u00e9 montr\u00e9e que le cortex cingulaire ant\u00e9rieur (CCA)<\/em> et ses circuits sont critiques pour les cons\u00e9quences anxiod\u00e9pressives de la douleur neuropathique. Les interneurones parvalbumine sont essentiels pour la balance excitation\/inhibition du cerveau et sont entour\u00e9s d\u2019un type de matrice extracellulaire particulier appel\u00e9 \u00ab\u00a0perineuronal nets\u00a0\u00bb. Les Perineuronal Nets assurent notamment un r\u00f4le de soutien aux neurones et limitent la plasticit\u00e9 des neurones. Le but de ma th\u00e8se est d\u2019\u00e9tudier le r\u00f4le des interneurones parvalbumine et leur perineuronal nets dans un mod\u00e8le murin de douleur chronique d\u00e9veloppant des sympt\u00f4mes anxiod\u00e9pressifs. C\u2019est dans ce contexte que j\u2019ai la chance de mener un projet sur la queue de l’aire tegmentale ventrale (tVTA)<\/em>, une structure mise en lumi\u00e8re, notamment par mon \u00e9quipe, il y a une quinzaine d\u2019ann\u00e9es. La tVTA, aussi appel\u00e9e noyau rostromedian du tegmentum (RMTg)<\/em> est une structure GABAergique, directement post\u00e9rieure aux noyaux dopaminergiques du m\u00e9senc\u00e9phale que sont l\u2019aire tegmentale ventrale et la substance noire compacte. La tVTA est consid\u00e9r\u00e9e comme le centre de contr\u00f4le inhibiteur majeur de ces noyaux dopaminergiques. Malgr\u00e9 une description anatomo-fonctionelle chez le rat, la tVTA reste peu d\u00e9crite chez la souris et difficile \u00e0 \u00e9tudier. \u00a0Mon projet de th\u00e8se vise donc \u00e0 identifier de nouveaux marqueurs de la tVTA, d\u2019en d\u00e9finir les limites anatomiques et le connectome chez la souris ainsi qu\u2019\u00e0 en caract\u00e9riser les r\u00f4les fonctionnels notamment dans l\u2019aversion et l\u2019activit\u00e9 locomotrice.<\/p>\t\t\t\t\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t\t<\/div>\n\t\t<\/div>\n\t\t\t\t\t<\/div>\n\t\t<\/section>\n\t\t\t\t
