repr\u00e9sente un co\u00fbt \u00e9conomique annuel de plusieurs milliards d&rsquo;euros en Europe. Ce type de douleur est la cons\u00e9quence directe d&rsquo;une l\u00e9sion ou d&rsquo;une maladie affectant le syst\u00e8me somatosensoriel. Les l\u00e9sions des nerfs sensoriels, ainsi que les changements p\u00e9riph\u00e9riques et centraux inadapt\u00e9s dans les r\u00e9seaux neuronaux qu\u2019ils contactent, conduisent \u00e0 l&rsquo;apparition et au d\u00e9veloppement de la douleur neuropathique. La pr\u00e9valence \u00e9lev\u00e9e de la douleur neuropathique en fait une priorit\u00e9 m\u00e9dicale qui reste tr\u00e8s difficile \u00e0 satisfaire en raison de  la complexit\u00e9 des syndromes concern\u00e9s et de l\u2019absence de strat\u00e9gies th\u00e9rapeutiques efficaces. \u00c0 ce jour, les traitements de premi\u00e8re intention comprennent des antid\u00e9presseurs ou des anticonvulsivants qui ne sont que mod\u00e9r\u00e9ment efficaces et seulement dans 60 % des cas. Dans les autres cas, des opiac\u00e9s ciblant les r\u00e9cepteurs opio\u00efdes mu sont prescrits. Cependant, l&rsquo;activation r\u00e9p\u00e9t\u00e9e de ces r\u00e9cepteurs induit des effets secondaires tels que la tol\u00e9rance qui n\u00e9cessite d&rsquo;augmenter la dose pour maintenir l&rsquo;effet analg\u00e9sique. Pour am\u00e9liorer l&rsquo;efficacit\u00e9 de ces traitements, il est crucial d&rsquo;identifier les alt\u00e9rations du contr\u00f4le opio\u00efde endog\u00e8ne qui le rendent incapable de pr\u00e9venir la douleur dans les conditions neuropathiques et qui contribuent \u00e0 r\u00e9duire l&rsquo;efficacit\u00e9 des traitements m\u00e9dicamenteux. Les m\u00e9canismes conduisant \u00e0 une r\u00e9ponse inadapt\u00e9e du syst\u00e8me opio\u00efde endog\u00e8ne dans les conditions neuropathiques repr\u00e9sentent donc des cibles th\u00e9rapeutiques potentielles. Dans ce contexte, notre projet vise sp\u00e9cifiquement \u00e0 identifier ces alt\u00e9rations afin de d\u00e9velopper des strat\u00e9gies th\u00e9rapeutiques plus efficaces. En outre, \u00e9tant donn\u00e9 la pr\u00e9valence plus \u00e9lev\u00e9e de la douleur neuropathique chez les femmes, l&rsquo;accent est \u00e9galement mis sur l&rsquo;identification des diff\u00e9rences li\u00e9es au sexe dans ces m\u00e9canismes. Pour r\u00e9aliser ces \u00e9tudes, notre \u00e9quipe allie mod\u00e8le murin et analyses depuis les niveaux mol\u00e9culaires et cellulaires jusqu\u2019aux niveaux int\u00e9gr\u00e9s en combinant approches biochimiques, imagerie de fluorescence \u00e0 haute r\u00e9solution, cultures primaires, tranches de cerveau et de moelle \u00e9pini\u00e8re, \u00e9lectrophysiologie, spectrom\u00e9trie de masse et comportement animal.\t\t\t\t\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t\t<\/div>\n\t\t<\/div>\n\t\t\t\t\t<\/div>\n\t\t<\/div>\n\t\t\t\t<div class=\"elementor-section elementor-top-section La balance excitation\/inhibition dans ce r\u00e9seau est essentielle pour moduler l\u2019information nociceptive. La plasticit\u00e9 \u00e0 court-terme des synapses inhibitrices de la lamina II, dans le cadre du traitement nociceptif, pourrait ajuster la sensibilit\u00e9 douloureuse. Gr\u00e2ce \u00e0 des enregistrements \u00e9lectrophysiologiques sur tranches de moelle \u00e9pini\u00e8re, j&rsquo;ai la possibilit\u00e9 de mettre en \u00e9vidence cette plasticit\u00e9 des synapses inhibitrices selon la nature excitatrice ou inhibitrice des neurones postsynaptiques. La morphine, utilis\u00e9e pour soulager les douleurs neuropathiques, est moins analg\u00e9sique compar\u00e9e \u00e0 son efficacit\u00e9 contre une douleur aigu\u00eb.
