neuroendocrines repose sur la s\u00e9cr\u00e9tion de mol\u00e9cules informatives, un processus r\u00e9gul\u00e9 par un intense trafic membranaire <em>(exocytose, endocytose, transport v\u00e9siculaire)<\/em>. <a href=\"https:\/\/inci.neuro.unistra.fr\/wp-content\/uploads\/2023\/03\/equipe1-figure1-300x300-1.jpg\"><img decoding=\"async\" class=\"alignright wp-image-5420\" d\u00e9crypter les m\u00e9canismes mol\u00e9culaires qui contr\u00f4lent ces diff\u00e9rentes \u00e9tapes de trafic et d\u2019en r\u00e9v\u00e9ler les alt\u00e9rations qui sont \u00e0 la base de pathologies importantes <em>(retard mental, maladies neurod\u00e9g\u00e9n\u00e9ratives, cancers neuroendocrines, ob\u00e9sit\u00e9\u2026)<\/em>.<\/p><p>En utilisant des neurones, des cellules neuroendocrines ou des cellules gliales, nous \u00e9tudions plus particuli\u00e8rement les m\u00e9canismes qui permettent \u00e0 diff\u00e9rents lipides et prot\u00e9ines de contr\u00f4ler l\u2019ensemble des \u00e9tapes de la vie des v\u00e9sicules, allant de la biog\u00e9n\u00e8se jusqu\u2019\u00e0 la fusion membranaire, en passant par leur transport au sein des \/>(doctorante)<\/figcaption><\/figure><p>Les bases mol\u00e9culaires de la s\u00e9cr\u00e9tion hormonale et de la neurotransmission ont longtemps \u00e9t\u00e9 \u00e9tudi\u00e9es \u00e0 travers le prisme des machineries prot\u00e9iques responsables de la fusion et du recyclage des v\u00e9sicules. Pourtant, la contribution des lipides membranaires \u00e0 ces processus reste largement sous-estim\u00e9e. Les phospholipides de la membrane plasmique sont distribu\u00e9s de mani\u00e8re asym\u00e9trique entre les deux feuillets de la bicouche, conf\u00e9rant \u00e0 la membrane une polarit\u00e9 structurale et fonctionnelle fondamentale. Nos travaux ont montr\u00e9 qu\u2019une perte locale et transitoire de cette asym\u00e9trie n\u2019est pas une simple cons\u00e9quence, mais une condition n\u00e9cessaire \u00e0 la s\u00e9cr\u00e9tion hormonale dans les cellules neuroendocrines et \u00e0 la neurotransmission dans des synapses \u00e0 haute fr\u00e9quence. Nous avons identifi\u00e9 la Scramblase-1 (PLSCR1) comme un r\u00e9gulateur central de ce processus. En effet, cette prot\u00e9ine perturbe localement l\u2019asym\u00e9trie lipidique pr\u00e8s des sites de fusion v\u00e9siculaire et r\u00e9gule le recyclage des v\u00e9sicules par endocytose compensatrice.<\/p><p>Mon projet vise \u00e0 d\u00e9crypter comment la PLSCR1 orchestre le couplage entre exocytose et endocytose, et \u00e0 \u00e9tablir comment les variations dynamiques de l\u2019asym\u00e9trie membranaire gouvernent le cycle exo-endocytose des v\u00e9sicules de s\u00e9cr\u00e9tion. Ces deux maladies impliquent une production excessive de peptides en mesure de s\u2019auto-assembler sous forme de fibres associ\u00e9es \u00e0 une certaine toxicit\u00e9 cellulaire. Il s\u2019agit des peptides amylo\u00efde-beta dans le cas d\u2019Alzheimer et de l\u2019amyline pour le diab\u00e8te de type 2. En r\u00e9alisant des \u00e9tudes de cin\u00e9tique de co-assemblage sur la formation de co-agr\u00e9gats, j\u2019\u00e9tudie l\u2019impact que peuvent exercer entre eux ces deux peptides. Des \u00e9tudes structurales permettent par ailleurs d\u2019observer et comparer les fibres form\u00e9es. L\u2019impact de ces co-agr\u00e9gats sur des cultures organotypiques d\u2019hippocampe de souris est \u00e9valu\u00e9 afin d\u2019en \u00e9tudier la toxicit\u00e9 par des approches immunocytochimique ainsi qu\u2019en mesurant l\u2019activit\u00e9 de r\u00e9seaux neuronaux par MEA.
