786w\" sizes=\"(max-width: 500px) 100vw, 500px\" \/><\/a>Une alimentation hypercaloriqie peut affecter l&rsquo;organisation circadienne de la prise alimentaire, entra\u00eenant un ph\u00e9nom\u00e8ne pathologique que nous avons appel\u00e9 \u00ab\u00a0chronob\u00e9sit\u00e9\u00a0\u00bb. Le noyau arqu\u00e9 de l&rsquo;hypothalamus <em>(ARC)<\/em> est une horloge alimentaire majeure dans le cerveau, situ\u00e9e \u00e0 la base du troisi\u00e8me ventricule et proche de l&rsquo;\u00e9minence m\u00e9diane. ARC est une porte d\u2019entr\u00e9e majeure pour les nutriments et les hormones circulantes et contient deux types principaux de cellules : les neurones exprimant le peptide li\u00e9 \u00e0 l&rsquo;agouti et le neuropeptide Y <em>(AgRP\/NPY)<\/em>, qui signalent la faim et stimulent la prise alimentaire, et les neurones exprimant la pro-opiom\u00e9lanocortine <em>(POMC)<\/em>, qui inhibent s\u00e9lectivement la prise alimentaire et limitent la prise de poids. D\u2019un point de vue neuroanatomique, ARC peut \u00eatre d\u00e9crit comme un centre int\u00e9grateur recevant d\u2019une part les signaux lumineux transmis par les noyaux suprachiasmatiques <em>(SCN)<\/em>, l&rsquo;horloge ma\u00eetresse du cerveau entra\u00een\u00e9e par la lumi\u00e8re, et d\u2019autre part les informations li\u00e9es \u00e0 l\u2019alimentation en provenance des organes p\u00e9riph\u00e9riques, relay\u00e9es par le nerf vague dans le noyau du tractus solitaire<em> (NTS)<\/em>. ARC interagit \u00e9galement et de mani\u00e8re importante avec d&rsquo;autres r\u00e9gions c\u00e9r\u00e9brales d\u00e9crites comme r\u00e9gulatrices du comportement alimentaire et de l&rsquo;app\u00e9tit, mais l&rsquo;organisation et la synchronisation de ce r\u00e9seau complexe dans le contexte de la chronob\u00e9sit\u00e9 restent largement m\u00e9connues. Dans notre laboratoire, nous \u00e9tudions ARC et ses interactions avec le corps et le cerveau chez des souris m\u00e2les et femelles, en r\u00e9ponse \u00e0 une d\u00e9r\u00e9gulation du rythme de prise alimentaire induite par une alimentation hypercalorique. En utilisant des approches de pointe telles que l&rsquo;enregistrement par photom\u00e9trie de fibre mono et multi-sites, le DREADD, la transparisation de tissus et le comportement chez le rongeur, nous cherchons \u00e0 comprendre comment ce r\u00e9seau complexe corps-cerveau peut \u00eatre perturb\u00e9 par un simple changement de r\u00e9gime alimentaire, en mettant particuli\u00e8rement l&rsquo;accent sur l&rsquo;implication de la composante h\u00e9patique des aff\u00e9rences p\u00e9riph\u00e9riques sensorielles.<\/p><p>En parall\u00e8le, afin de d\u00e9tecter les diff\u00e9rentes r\u00e9ponses du SCN et des deux horloges entra\u00een\u00e9es par la nourriture<em> (ARC et cervelet)<\/em> aux synchroniseurs nutritionnels, nous \u00e9tudierons in vitro les effets chronomodulateurs des lipides et des sucres sur des explants organotypiques de souris PER2::Luc. Ensuite, nous ph\u00e9notyperons les cellules du SCN, de ARC et du cervelet impact\u00e9es \u00e0 l&rsquo;aide d&rsquo;un Luminoview.<\/p>\t\t\t\t\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t\t<\/div>\n\t\t<\/div>\n\t\t\t\t\t<\/div>\n\t\t<\/div>\n\t\t\t\t<div class=\"elementor-section elementor-top-section elementor-element
