876w\" sizes=\"(max-width: 450px) 100vw, 450px\" \/><\/a>Dans ce projet<\/strong>, nous \u00e9valuons l&rsquo;impact des perturbations du cycle lumi\u00e8re\/obscurit\u00e9 sur les fonctions m\u00e9taboliques et reproductives, en particulier chez les rongeurs femelles. Ce projet implique \u00e0 la fois les souris nocturnes conventionnelles chez lesquelles les m\u00e9canismes neuroendocriniens r\u00e9gulant la reproduction et le m\u00e9tabolisme quotidiens sont bien connus, et les rats roussards diurnes <em>(Arvicanthis)<\/em> en tant que nouveau mod\u00e8le animal de physiopathologie humaine.<br \/>Nous avons d\u00e9j\u00e0 montr\u00e9 que les rats roussards m\u00e2les expos\u00e9s \u00e0 3 mois de d\u00e9calage horaire hebdomadaire pr\u00e9sentent une intol\u00e9rance au glucose, un raccourcissement des t\u00e9lom\u00e8res (marqueur cellulaire du vieillissement biologique) et une diminution de l&rsquo;expression h\u00e9patique de SIRT1, qui une d\u00e9sac\u00e9tylase d\u00e9pendante du NAD+ qui module l&rsquo;horloge mol\u00e9culaire et prot\u00e8ge les t\u00e9lom\u00e8res du raccourcissement <em>(Grosbellet et al. FASEB J, 2015)<\/em>. Ici, notre objectif est de distinguer l&rsquo;impact direct de la lumi\u00e8re la nuit de l&rsquo;impact de la d\u00e9synchronisation du syst\u00e8me circadien, et de comprendre comment les perturbations circadiennes peuvent acc\u00e9l\u00e9rer le vieillissement cellulaire et \u00eatre diab\u00e9tog\u00e8nes chez la femme. Tout d&rsquo;abord, nous effectuerons des tests fonctionnels pour le m\u00e9tabolisme et la reproduction chez des femelles de rat roussard expos\u00e9es \u00e0 un d\u00e9calage chronique du cycle lumi\u00e8re\/obscurit\u00e9 ou \u00e0 de la lumi\u00e8re la nuit. Ensuite, la d\u00e9sorganisation circadienne interne sera \u00e9valu\u00e9e par l&rsquo;analyse des profils quotidiens d&rsquo;expression des g\u00e8nes d\u2019horloge sur des \u00e9chantillons hypothalamiques et p\u00e9riph\u00e9riques. En outre, l&rsquo;expression de g\u00e8nes sp\u00e9cifiques du m\u00e9tabolisme et de la reproduction sera analys\u00e9e.<br \/>Nous \u00e9tudierons \u00e9galement les voies intracellulaires impliqu\u00e9es dans le vieillissement au sein du foie, du pancr\u00e9as et du SCN. En fonction des voies de signalisation les plus affect\u00e9es, nous tenterons d\u2019emp\u00eacher le vieillissement cellulaire par des traitements pharmacologiques cibl\u00e9s. Nous d\u00e9terminerons \u00e9galement si les r\u00e9ponses \u00e0 l&rsquo;insuline sont augment\u00e9es ou diminu\u00e9es pendant le test de tol\u00e9rance au glucose afin de dissocier la r\u00e9sistance \u00e0 l&rsquo;insuline d\u2019un d\u00e9ficit de synth\u00e8se d\u2019insuline. Nous avons r\u00e9cemment montr\u00e9 que l&rsquo;exposition \u00e0 un d\u00e9calage horaire chronique abolit le pic pr\u00e9ovulatoire de LH et r\u00e9duit la fertilit\u00e9 de souris femelles. Comme un nombre croissant de femmes travaillent en horaires d\u00e9cal\u00e9s, il est important de mieux comprendre les m\u00e9canismes impliqu\u00e9s afin de proposer des strat\u00e9gies pour limiter les cons\u00e9quences n\u00e9gatives du travail post\u00e9 sur la reproduction. Les objectifs de ce projet sont:<\/p><ol><li>d&rsquo;analyser comment les rythmes journaliers et estraux de l&rsquo;axe hypothalamo-pituitaire-ovarien sont alt\u00e9r\u00e9s en cas de travail post\u00e9 chronique ;<\/li><li>d&rsquo;\u00e9valuer si l&rsquo;exposition au travail post\u00e9 chronique alt\u00e8re la fertilit\u00e9 des femelles et le d\u00e9veloppement de leur prog\u00e9niture ;<\/li><li>d&rsquo;\u00e9tablir des protocoles pour restaurer cette fertilit\u00e9; et<\/li><li>en collaboration avec le service d&rsquo;endocrinologie des H\u00f4pitaux de Strasbourg <em>(financements Idex Unistra &amp; SFD)<\/em>, de d\u00e9terminer si les femmes travaillant en horaires d\u00e9cal\u00e9s pr\u00e9sentent \u00e9galement des alt\u00e9rations de leurs index reproductifs et m\u00e9taboliques.<\/li><\/ol>\t\t\t\t\t\t\t\t<\/div>\n\t\t\t\t<\/div>\n\t\t\t\t\t<\/div>\n\t\t<\/div>\n\t\t\t\t\t<\/div>\n\t\t<\/div>\n\t\t\t\t<section class=\"elementor-section
