de l&rsquo;hormone pin\u00e9ale, la m\u00e9latonine, est traduit en une augmentation en \u00e9t\u00e9\/longue photop\u00e9riode de la synth\u00e8se de TSH <em>(thyroid stimulating hormone)<\/em> par la pars tuberalis de l&rsquo;hypophyse. En collaboration avec le Dr H. Dardente <em>(INRA, Nouzilly ; ANR GMO-Phen)<\/em>, nous testons l&rsquo;hypoth\u00e8se d&rsquo;un r\u00f4le central des neurones RFRP3 dans l&rsquo;adaptation saisonni\u00e8re de la reproduction. Chez le hamster syrien et le mouton, nous d\u00e9veloppons la technologie Crispr-Cas9 pour \u00e9valuer l&rsquo;effet du knock-out de RFRP3 sur la reproduction saisonni\u00e8re, et nous comparons les changements photop\u00e9riodiques dans le transcriptome des neurones RFRP3.<\/p><p><strong>Hibernation<\/strong>. Les hamsters syriens maintenus dans une photop\u00e9riode courte \u00e0 basse temp\u00e9rature sont capables d&rsquo;hiberner. Nous \u00e9lucidons les voies neuroendocriniennes impliqu\u00e9es dans le d\u00e9clenchement et l&rsquo;arr\u00eat des cycles d&rsquo;hibernation en collaboration avec D. Hazlerigg et S. Wood de l&rsquo;Universit\u00e9 arctique de Tromso<em> (Norv\u00e8ge ; financements INSB Diversity in Biological Mechanisms)<\/em>. Les donn\u00e9es pr\u00e9liminaires montrent que la TSH, perfus\u00e9e icv via des minipompes osmotiques programmables, emp\u00eache les \u00e9pisodes d&rsquo;hypothermie. De plus, dans des zones hypothalamiques sp\u00e9cifiques, des cellules qui restent \u00e0 identifier sont activ\u00e9es \u00e0 diff\u00e9rents stades du cycle d&rsquo;hibernation.<\/p><p><strong>Programmation maternelle saisonni\u00e8re<\/strong>. La m\u00e9latonine maternelle est capable de programmer le d\u00e9veloppement de la prog\u00e9niture, puisque des hamsters n\u00e9s d&rsquo;une gestation maternelle sous photop\u00e9riode longue ont un d\u00e9veloppement m\u00e9tabolique et reproductif plus rapide que les hamsters n\u00e9s d&rsquo;une gestation maternelle sous photop\u00e9riode courte. En collaboration avec S. Wood et d. Hazlerigg de l&rsquo;Universit\u00e9 Arctique de Tromso <em>(Norv\u00e8ge)<\/em>, nous \u00e9tudions le r\u00f4le de la plasticit\u00e9 des tanycytes dans cette programmation maternelle saisonni\u00e8re m\u00e9di\u00e9e par la m\u00e9latonine.<\/p><p><strong>Neurogliogen\u00e8se saisonni\u00e8re<\/strong>. L&rsquo;adaptation saisonni\u00e8re implique d&rsquo;importants changements dans la plasticit\u00e9 des cellules hypothalamiques et l&rsquo;expression des g\u00e8nes. Le transfert en photop\u00e9riode courte est associ\u00e9 \u00e0 une augmentation de la neuro-gliogen\u00e8se hypothalamique. Nous \u00e9tudions les signaux saisonniers r\u00e9gulant cette prolif\u00e9ration cellulaire induite par photop\u00e9riode courte, le devenir des cellules prolif\u00e9ratives et le r\u00f4le putatif de ces nouvelles cellules hypothalamiques dans les fonctions
